Abstract: OBJECTIVE To establish a characteristic fingerprints method for qualitative analysis of the main components in total flavonoids extracts of Daidai in vitro and in vivo by UPLC-MS,laying a foundation for the research of components in plasma and the clarifying the material basis of total flavonoids extracts of Daidai.METHODS The characteristic fingerprints of total flavonoids extracts of Daidai were established by UPLC-MS,the characteristic fingerprints of plasma of normal and hyperlipidaemia rats were compared after oral administration of total flavonoids extracts of Daidai.RESULTS The conditions of chromatography and mass spectrometry were optimized and the best blood sampling time and plasma pretreatment method were determined;RSDs of relative retention time of 9 targeted peaks were all below 0.5%;RSDs of relative peak area were all below 2.9%;extracting recovery were all over 75%.The characteristic fingerprints of total flavonoids extracts of Daidai were established in vitro and in vivo.CONCLUSION The established characteristic fingerprints in vivo and in vitro of total flavonoids extracts of Daidai were stable and accurate,which could be used to the components analysis in vivo and in vitro of normal and pathological state,providing beneficial exploration for‘characteristic fingerprint of activity’.
