Efficacy of Osmotic Pressure and Plant Growth Regulators in Media on Conservation of Grape Plantlets in vitroChinese Full Text

Abstract: In order to seek a way to prolong the subculture interval during grape germplasm conservation, the effects of osmotic pressure and plant growth regulators in culture media on growth of Melissa, Jumeigui, Sweet Scarlet and Heimeigui grape plantlets in vitro was studied. The osmotic pressure was adjusted by supplying sugar or mannitol with different concentration in culture media. The plant growth regulators used in this experiment were PP333, CCC, or ABA. The results showed that 20~30 g/L mannitol or 2~3 mg/L PP333 in the culture media could significantly inhibit the plantlet growth with a high survival percentage in Melissa and Jumeigui grapes,which was suitable for retardant growth subculture during conservation in vitro. No significant inhibit growth efficacy found in Melissa and Sweet Scarlet grapes with 25~85 g/L sugar and in Melissa and Sweet Scarlet grapes with 300 mg/L or less CCC in culture media. The plantlets showed vitrification phenomenon when 400 mg/L or more CCC was supplied. 3 mg/L or more ABA in culture media did not inhibit the growth of Jumeigui and Heimeigui grapes, and that could lead to the death of the plantlets as well. CCC or ABA in culture media was not suitable for retardant growth subculture for these applied grape cultivars.
