Effect of VEGF on proliferation and migration ability of human umbilical vein endothelial cells cultured in vitroChinese Full Text

Abstract: AIM To explore the effect of vascular endothelial growth factor (VEGF) on proliferation and migration ability of human umbilical vein endothelial cells (HUVECs) cultured in vitro. METHODS The HUVECs were transfected by VEGF gene and the VEGF protein was added into the culture fluid for HUVECs. The expression of VEGF mRNA was detected by RT-PCR to evaluate the efficacy of the transfection. The effect of VEGF on the proliferation and migration ability of HUVECs were measured by 3H-thymidine incorporation and by scarification test,respectively. RESULTS The result of RT-PCR showed that after transfecting VEGF gene into HUVECs by lipidosome,the expression of VEGF mRNA in HUVECs was higher than that in control group,suggesting that the transfection of VEGF gene was successful. The results of 3H-thymidine incorporation and scarification test indicated that transfecting VEGF gene and adding the VEGF protein into culture fluid could promote the proliferation and migration ability of cultured HUVECs. CONCLUSION VEGF promotes the proliferation and migration of cultured HUVECs. Transfecting the VEGF gene into HUVECs and adding the VEGF protein into the culture fluid for HUVECs have the same effect.
