Effects of EMP on morphology of rat’s myocardial intercalated discs and the expression of Zo-1 and Cx43 proteinsChinese Full Text

Abstract: AIM To explore the mechanism of intercalated discs (ID) caused by electromagnetic pulse (EMP) radiation so as to provide experimental basis for establishing sanitary standard and its medial application. METHODS EMP (200 kv/m field intensity, 200 pulse and 2 s pulse interval from EMP simulator) was used in 54 female SD rats. The rats were randomly divided into seven groups, 12 rats in control group and 12 h group, and 6 rats in the other 5 groups (immediately, 6, 24, 36 and 48 h group). At different time points after EMP radiation (immediately, 6, 12, 24, 36 and 48 h), lanthanum nitrate tracer method and the transmission electron-microscope (TEM) were used to observe the structural changes of left ventricular myocardium of the rats. At 12 h after EMP radiation, the changes of gap junctions Zo-1 and gap junctions Cx43 were observed by fluorescent labeling and laserconfocal microscopy. RESULTS No obvious changes were observed under TEM in cardiac muscles of the rats after EMP radiation. The change in ID of cardiac muscle was most obvious at 12 h after radiation and a large amount of lanthanum nitrate. The change in ID of cardiac muscle was less obvious at 36 h than that at 12 h and only a small amount of lanthanum nitrate was seem. At 48 h after radiation, it became almost normal and no lanthanum nitrate was seen. Gap junctions Zo-1 and gap junctions Cx43 expressed at the cardiac muscle cell conjunction in control and 12 h group, but expression tended to decrease in radiation groups. CONCLUSION After EMP exposure, the broadening of space between intercalated discs changes along with time. The changes in ID of cardiac muscle have reversibility by themselves. The expressions of gap junctions Zo-1 and gap junctions Cx43 have a downtrend.
