Abstract: This study was aimed at investigating the changes of nitric oxide(NO) concentration in vitro vascular smooth muscle cells(VSMC) of essential hypertension patients(EH) and the role of these changes in the pathogenesis of hypertension. In our study, the subjects were divided into two groups, the EH group, and the normal control group. We seperated and cultured VSMC of the two groups by the compound collagenase enzyme method. In order to observe the character of EH NO secretion, we examed the NO concentration and the activity of NOS in VSMC. The results showed as follows:①The NO concentration and the activity of NOS in VSMC of the EH group were significantly lower than those of the control group( P <0.01);②The NO concentration and the activity of NOS in VSMC of both the groups increased with the prolonged cultivation time;③The two groups we observed that the concentration of NO was increasing with the increasing activity of enzyme. And there was a positive correlation between the NO concentration and the activity of enzyme. Our study demonstrates that the NOS NO system of EH is probably abnormal in structure or function and is involved in the activation and development of essential hypertension.
