摘要:
目的:研究萝卜硫素对大鼠胃癌组织中相关基因表达的影响。方法:对萝卜硫素组大鼠灌胃50 mg/kg萝卜硫素,胃癌组大鼠腹腔注射同体积生理盐水,采用基因芯片筛选出萝卜硫素诱导表达差异的基因,利用Real-time PCR方法检测胰岛素样生长因子结合蛋白3(insulin-like growth factor binding protein 3,IGFBP3)、神经纤维蛋白1(Neuropilin-1)、雄性性别决定基因相关高迁移率族盒基因(sex-determining regio of Y chromosome related high-mobility-group box 4,SOX4)、N-myc下游调节基因1(NDRG1)4个基因水平;Western Blot方法检测IGFBP3、NDRG1、SOX4和Neuropilin-1蛋白表达水平。结果:经萝卜硫素治疗后大鼠胃癌组织表达上升的基因为159个(差异倍数>2.0),表达下降的基因有67个(差异倍数<0.5)。这些基因主要参与免疫调节、抗氧化、抗炎、介导细胞增殖及凋亡等生物过程。Real-time PCR发现IGFBP3和NDRG1基因表达明显上调,SOX4和Neuropilin-1基因表达明显下调(P<0.05),结果与基因芯片一致。Western Blot实验证实萝卜硫素治疗后仅IGFBP3蛋白表达升高(P<0.05),其他3种蛋白表达没有显著差异(P>0.05)。结论:基因芯片与Real-time PCR 技术相结合可找出萝卜硫素处理后的差异表达基因,可为探讨萝卜硫素抑制胃癌的作用机制提供依据。
Abstract:
Objective: To study the effects of sulforaphane on the related gene expressions in gastric cancer tissue of rats. Methods: The rats in sulforaphane group were drenched with 50 mg/kg of sulforaphane, the rats in gastric cancer group accepted intraperitoneal injection of the same volume of saline, gene chips was used to screen the differential expressions genes induced by sulforaphane, Real-time PCR method was adopted to determine the levels of IGFBP3, Neuropilin-1, SOX4 and NDRG1; Western blot to analyze the levels of IGFBP3, NDRG1, SOX4 and Neuropilin-1 protein. Results: After treated by sulforaphane, 159 genes showed up-regulated expression in gastric cancer tissue of the rats (the fold change>2.0), 67 genes down-regulated expression (the fold change<0.5). These genes were mainly involved in biological courses such as immune regulation, anti-oxidation effects, anti-inflammation, mediating cellular proliferation and apoptosis. Real-time PCR suggested that genetic expressions of IGFBP3 and NDRG1 up-regulated, while genetic expressions of SOX4 and Neuropilin-1 down-regulated notably(P<0.05), and the results were identical to gene chip. Western blot suggested that only the expressions of IGFBP3 protein rose apparently after sulforaphane treatment (P<0.05), no significant difference was found in the expressions of the other three proteins(P>0.05). Conclusion: Gene chip combined with Real-time PCR technology could find out the differential expressions gene after sulforaphane management, and it could provide the reference for the discussion on the mechanism of sulforaphane inhibiting gastric cancer.
