Changes in the Adenylate Energy Charge in Human Blood Platelets induced by Adenosine Diphosphate

Abstract

WHEN platelet rich plasma at 37° C is exposed to low concentrations of adenosine diphosphate (ADP) the platelets change in shape from flat disks to irregular spheres1, and if the mixture is stirred and provided that necessary cofactors including calcium ions and fibrinogen are present, aggregation then takes place. The detailed mechanism of this response to ADP is the subject of a number of speculative hypotheses2–5. Here I report an observation on the effects of exogenous ADP on platelet adenine nucleotide metabolism that may be relevant to the induction of platelet aggregation.
