Synthesis of adenophostin A

Abstract

The natural product and potent agonist of the d-myo-inositol 1,4,5-trisphosphate receptor, adenophostin A, was synthesised from adenosine and d-glucose using efficient methodology. The synthetic material was equipotent with naturally occurring adenophostin A in evoking Ca2+ release from the intracellular stores of permeabilised cells.

      D-myo-inositol 1,4,5- trisphosphate (IP3) was from Enzo (Exeter, UK). Synthetic adenophostin A [24] was a generous gift from Prof Barry V. L. Potter, University of Oxford. Mag-Fluo-4 AM, Mag-Fluo-4 tetrapotassium salt, rabbit Fluorescein/Oregon Green polyclonal antibody (QAb, catalogue number A-889), water-soluble probenecid, RPMI medium and Dulbecco's modified Eagle’s medium/nutrient mixture F-12 with GlutaMAX (DMEM/F-12 GlutaMAX) were from ThermoFisher Scientific (Paisley, UK).

      The result suggested the existence of an internal hydrogen-bonding structure between the oxygen atom of the P = O and one of the hydrogen atoms of the amino group in the guanine base. IMT was uniquely employed for phosphitylations of three secondary hydroxy groups of the adenine-unprotected intermediate 8 derived from adenosine and glucose (Scheme 3).24 Subsequent oxidation with m-CPBA followed by hydrogenolysis of the benzyl groups afforded adenophostin A, a potent D-myo-inositol 1,4,5-triphosphate receptor-agonist discovered from a culture broth of Penicillium brevicompactum.25,26

      Many different conformations of adenophostin are therefore possible and the receptor-bound conformation is difficult to predict. Molecular modeling suggests that adenophostin can adopt extended conformations in which the adenosine holds the 2′-phosphate close to the glucose ring in a position similar to that occupied by the 1-phosphate of IP3 (Fig. 1B; Hotoda et al., 1999; Marwood, Correa, Taylor, & Potter, 2000). NMR studies of adenophostin in solution also support some aspects of this type of model (Felemez, Marwood, Potter, & Spiess, 1999; Hotoda et al., 1999).
