A Polygalacturonase-Inhibiting Protein in Alfalfa Callus Cultures

Summary

The activity of a protein (PGIP) that inhibits fungal polygalacturonase was assayed in calli of alfalfa (Medicago sativa L). PGIP activity in calli was higher than that measured in hypocotyls and leaves of alfalfa plants. Medicago PGIP was purified by affinity chromatography through Sepharose 4B covalently linked to homogeneous Aspergillus niger polygalacturonase. Purified PGIP was a basic protein and its isoelectric point was similar to that reported for Phaseolus vulgaris PGIP (Cervone et al., 1987b).
