Abstract

A nine-step synthesis of 9-amino-4-methoxypyrido[3′,2′:4,5]pyrrolo[1,2-c]pyrimidine, a tricyclic ring system present in the marine alkaloids variolins is described. The natural marine products meridianins C–E have been synthetized for the first time starting from N-protected 3-acylindoles.

Protecting-Group-Free Synthesis of Meridianin A-G and Derivatives and Its Antibiofilm Evaluation

Herein, a protecting-group-free protocol was developed to realize a time and step economy diversification of the Meridianin alkaloid. A broad range of substituents are tolerated to deliver the products in moderate to high yields, and the first synthesis of Meridianin B was achieved. The simplicity of this protocol enables the rapid construction of a Meridianin derivative library for antibiofilm evaluation. Preliminary results reveal that Meridianin derivatives were capable of inhibiting the Acinetobacter baumannii biofilm and lowering the antibiotic MIC synergistically.
Recent developments in the synthesis of 4-, 5-, 6- and 7-azaindoles

Azaindole structures, also named as pyrrolopyridines, have gained significant relevance in the last three decades. Many different synthetic approaches have been proposed due to their structural relation with the indole moiety. The outstanding development in the synthesis of heterocycles and also, in cross-coupling reactions, has provided a wide range of synthetic approaches to azaindoles. Azaindoles are extremely relevant in different scientific domains: drug research, natural products, photochemistry, etc. The purpose of this review is to structure all these methods with a retrosynthetic approach inspired by indole (pyrrole) or quinoline (pyridine) synthesis in order to provide an overall perspective of how to attempt the preparation of these heterocycles.

From a synthetic point of view, 2-aminopyrimidines can be obtained by condensation reactions between α,β-unsaturated carbonyls and guanidine [10]. Indeed, the intermediates in the synthesis of meridianin alkaloids C (1), D (2) and E (3) (see Fig. 1) were synthesized by the previously mentioned reaction in yields of 72-82% [11]. Additionally, 2-aminopyrimidines can be synthesized by the condensation of chalcones with guanidine.
The pyrimidine core is present in a wide variety of compounds that show interesting fluorescent properties and have been used as pigments and dyes. In this research, we synthesized 4-(4-(dimethylamino)phenyl)-6-phenylpyrimidin-2-amine (compound 9) from chalcone (8) with 45% yield. We obtained monocrystals of 9 by slow evaporation of dichloromethane as a solvent. Additionally, we measured the photophysical properties of compound 9, and its absorption wavelength (λ_abs), molar absorption coefficient (ε), excitation wavelength (λ_exc) and emission wavelength (λ_em), as well as Stokes shift (Δλ), increase with increasing solvent polarity. These properties are all related to the stabilization of the excited state by the solvent. On the other hand, the quantum yields (Φ values) in toluene and CH₂Cl₂ were notable greater than those in other solvents due to their high optical density (OD) and the decrease in the loss of fluorescence by non-radiative processes
Novel meriolin derivatives as rapid apoptosis inducers

Quite early 3-(hetero)aryl substituted 7-azaindoles have therefore become a lead structure for the development of kinase inhibitors. Indeed, meriolins have been reported to strongly inhibit various protein kinases.5,6 This finding correlates excellently with the ability of 7-azaindoles to bind to the hinge region of kinases.
3-(Hetero)aryl substituted 7-azaindoles possessing multikinase inhibitor activity are readily accessed in a one-pot Masuda borylation-Suzuki coupling sequence. Several promising derivatives were identified as apoptosis inducers and, emphasizing the multikinase inhibition potential, as sphingosine kinase 2 inhibitors. Our measurements provide additional insights into the structure-activity relationship of meriolin derivatives, suggesting derivatives bearing a pyridine moiety with amino groups in 2-position as most active anticancer compounds and thus as highly promising candidates for future in vivo studies.
Chemical Synthesis of Meridianins and Related Derivatives

The meridianins are alkaloids characterized by an indole framework bearing a 2-aminopyrimidine ring at the 3-position. These structures are emerging as valuable pharmacophores following their discovery in the late 1990s and exhibit a wide range of potent biological activities. Modification of derivatives have been a fruitful endeavor and lead to new and/or improved biological activities. Syntheses of these targets have developed from the step-inefficient Bredereck protocol, to a more elegant focus on transition metal catalysis. The disclosed review concerns the synthesis of meridianins and related structures.
Pim kinase inhibitory and antiproliferative activity of a novel series of meridianin C derivatives

A novel series of meridianin C derivatives substituted at C-5 position were prepared. These derivatives were tested for their kinase inhibitory potencies against all three family members of the pim kinases (Pim-1, Pim-2 and Pim-3). In addition, their antiproliferative activity towards three human leukemia cell lines as MV4-11, Jurkat clone E6-1 and K562 has been evaluated. Structure activity relationships at C-3 and C-5 positions of indole were performed to better understand the mechanism behind the enhanced potency. Compound 7f, the most active compound of the series showed a single-digit nanomolar IC₅₀ with selectivity towards Pim-1 kinase.
