2016, Journal of Biological ChemistryCitation Excerpt :This gabapentin concentration is close to that in patient cerebrospinal fluid after chronic gabapentin treatments. It has been reported that oral 900 mg/day gabapentin treatment for 3 months can reach peak plasmid concentration ∼10 mg/liter (58 μm) 3 h after the last dose (65), and cerebrospinal fluid gabapentin concentration range was ∼20–35% that in the plasma after multiple dosing (65, 66). Collectively, these results support the idea that TSP4/Cavα2δ1-dependent processes mediate both TSP4-induced pain states and the putative pathological underpinning (increased mEPSC frequency).

Furthermore, gabapentin [4] does not increase GABA in human cerebrospinal fluid like vigabatrin [3]. Therefore, gabapentin and pregabalin are not GABAergic drugs, and they have no actions or molecular targets unique to GABA synapses.
