Peroxynitrite concentration was determined spectrophotometrically at 302 nm (ϵ302 nm = 1670 m−1 cm−1) (23). The spin trap 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) and the anti-DMPO rabbit antiserum were kindly provided by Dr. Ronald Maison (NIEHS, National Institutes of Health, Bethesda, MD).

However, nitric oxide reacts with superoxide at a high rate constant to give peroxynitrite (ONOO−), able to decompose spontaneously to yield nitrogen dioxide (•NO2) and hydroxyl radicals [2]. Under normal circumstances, the formation and subsequent degradation of reactive species is regulated by cellular defense systems, including scavenging enzymes able to remove oxidants or their precursors such as superoxide dismutases, catalase, thioredoxin/thioredoxin reductase, and glutathione peroxidase.

2005, Methods in EnzymologyCitation Excerpt :A simpler method of synthesis was first performed in 1902 when Baeyer and Villiger (1901) published a method of mixing nitrite with hydrogen peroxide. In 1968, other authors (Hughes and Nicklin, 1968, 1970a,b; Hughes et al., 1971) reported a synthesis also based on the reaction between nitrite and hydrogen peroxide in which the hydrogen peroxide was “thrown” into a nitrite solution and produced peroxynitrite in yields between 40 and 50%. In 1973, Reed et al. (1974) reported a method of reacting nitrite and hydrogen peroxide in a quenched flow reactor.
