Genetic variation, hippocampal mossy fibres distribution, novelty reactions and spatial representation in mice

Abstract

It is now well established that variation in hippocampal circuitry correlates with various behavioural measures. In order to formulate more precise hypotheses about the functional role of the supra (SP-MF) and intra-infra mossy fibres (IIP-MF) layers, we investigated their covariation with a large number of behavioural measures in the classic and the spatial open-field taken from a genetically heterogeneous population composed of subjects from 12 isogenic mice groups (9 inbred strains and 3 F1's). The hippocampus-behaviour intercorrelation matrix reveals that behavioural reactions and inferred psychological processes only correlate with IIP-MF. Detailed analysis of correlates shows that IIP-MF positively correlate with locomotor activity, rearings and central sector crossings in the classic open-field and correlate negatively with the short-term habituation index to the objects in the spatial open-field. Surprisingly the memory of the spatial arrangement of a set of objects does not correlate with any hippocampal measure.

    Less than 10 years later, it has been adapted to mice (Dodart et al., 1997; Messier, 1997), and more recently to other species (Languille et al., 2015; Ardiles et al., 2012).     These behavioral results support the latest findings in which functional inactivation of dorsal CA2 caused no deficit in hippocampus-dependent spatial memory [32]. However, given the previously reported involvement of the hippocampus and its general modulatory role in rearing behavior [13,54,5,31,12,29,62] regional structural alterations, especially in dorsal CA2, highlight the pivotal role of the CA2 region in such behaviors that commonly appear independent of the hippocampus.     The hippocampal MF comprise the axons of the dentate gyrus granule neurons that innervate the CA3 field (Gaarskjaer, 1986). Morpho-behavioral correlations in genetically-altered mice implicate hippocampal MF with exploratory behavior (Roullet and Lassalle, 1990; Ivanco and Greenough, 2002; Mineur et al., 2002).     For example, we recently showed how neonatal FGF2 treatment shifts the developmental trajectory of the hippocampus, leading to a life-long change in neurogenesis, hippocampal morphology and gene expression (Turner et al., 2011). Considering the myriad roles of the hippocampus in guiding emotional behavior and exploration of the environment, which are certainly relevant to addiction-related behavior (Bannerman et al., 2002; Gray, 1982; Jacobson and Sapolsky, 1991; Jeewajee et al., 2008; Lever et al., 2006; McNaughton and Gray, 2000; Roullet and Lassalle, 1990; Sapolsky et al., 1984), we cannot exclude the possibility that increased cocaine sensitization in neonatal FGF2-treated bLRs is linked to changes in their hippocampus. Future experiments are warranted to explore these possibilities.
