New method for the quaternization of chitosan
Abstract
Trimethyl chitosan ammonium iodide was obtained by reaction of a low acetyl content chitosan with methyl iodide and sodium hydroxide under controlled conditions. The role of sodium iodide as an electrostatic charges screening salt is discussed. The reaction was performed in several steps to obtain derivatives of chitosan at various degrees of quaternization with a limit value near 64%. For a degree of quaternization greater than 25%, these polymers are soluble in water, whatever the pH.

Optimizing N,N,N-trimethyl chitosan synthesis: A design of experiments (DoE) approach
2024, Carbohydrate PolymersThis study aimed to optimize the synthesis of trimethyl chitosan (TMC) with a high degree of N,N,N-trimethylation (DTM) through a one-step procedure, minimizing reagent use, reaction time, and avoiding O-methylation, using the Design of Experiments (DoE) approach. Initially, sequential designs were done. Following the determination of the initial conditions a Fractional Factorial Design was used, investigating methyl iodide (MeI) and NaHCO3 molar ratios, temperature, and reaction time on DTM. MeI and NaHCO3 molar ratios were found to be significant (p-values equal to 0.02 and 0.02, respectively), the reaction temperature (p = 0.04) displayed a non-linear effect, while the reaction time was found to be non-significant (p = 0.93). Finally, a Full Factorial Design was done to optimize temperature and base addition methods. Incremental addition of the base was determined to be feasible without affecting the DTM, thereby preventing any viscosity-related problems. DTM was achieved up to 72 % in a one-step procedure, with no O-methylation. These optimized conditions offer a cost-effective, one-step synthesis method for TMC production, holding significant promise for industrial applications by avoiding multistep reactions, ensuring minimal reagent use, and preventing O-methylation. The findings mark a substantial advancement in TMC synthesis, presenting a streamlined and efficient approach with substantial practical implications for process development.
Quaternary-ammonium chitosan, a promising packaging material in the food industry
2024, Carbohydrate PolymersQuaternary-ammonium chitosan (QAC) is a polysaccharide with good water solubility, bacteriostasis, and biocompatibility. QAC is obtained by methylating or grafting the quaternary-ammonium group of chitosan and is an important compound in the food industry. Various QAC-based complexes have been prepared using reversible intermolecular interactions, such as electrostatic interactions, hydrogen bonding, metal coordination, host-guest interactions, and covalent bonding interactions consisting of Schiff base bonding and dynamic chemical bond cross-linking. In the food industry, QAC is often used as a substrate in film or coating for food preservation and as a carrier for active substances to improve the encapsulation efficiency and storage stability of functional food ingredients. In this review, we have assimilated the latest information on QAC to facilitate further discussions and future research. Advancement in research on QAC would contribute toward technology acceleration and its increased contribution to the field of food technology.
Chitosan-based hemostatic sponges as new generation hemostatic materials for uncontrolled bleeding emergency: Modification, composition, and applications
2023, Carbohydrate PolymersThe choice of hemostatic technique is a curial concern for surgery and as first-aid treatment in combat. To treat uncontrolled bleeding in complex wound environments, chitosan-based hemostatic sponges have attracted significant attention in recent years because of the excellent biocompatibility, degradability, hemostasis and antibacterial properties of chitosan and their unique sponge-like morphology for high fluid absorption rate and priority aggregation of blood cells/platelets to achieve rapid hemostasis. In this review, we provide a historical perspective on the use of chitosan hemostatic sponges as the new generation of hemostatic materials for uncontrolled bleeding emergencies in complex wounds. We summarize the modification of chitosan, review the current status of preparation protocols of chitosan sponges based on various composite systems, and highlight the recent achievements on the detailed breakdown of the existing chitosan sponges to present the relationship between their composition, physical properties, and hemostatic capacity. Finally, the future opportunities and challenges of chitosan hemostatic sponges are also proposed.
Coating of surgical masks with quaternized chitosan aiming at inactivating coronavirus and antibacterial activity
2023, Carbohydrate Polymer Technologies and ApplicationsSARS-CoV-2 caused a global pandemic with high transmission rates, even with widespread vaccination. In addition to isolation and prevention measures, it is important to find new solutions to inactivate the virus and prevent its viability on surfaces, such as clothing and face masks. Chitosan derivatives, with their antibacterial activity, biocompatibility, and non-toxicity, are a promising option for coating these components with virucidal materials. N-(2-hydroxypropryl)-3-trimethylammonium chloride, a quaternized chitosan derivative with degrees of substitution ranging from 59 to 93%, was synthesized. This derivative was used to coat cotton and polypropylene fabrics, such as face masks, without visibly affecting their morphology or filtering capacity. In vitro tests demonstrated that the coatings produced had high antiviral activity against coronavirus, with efficacy up to 99.99%, and were also bactericidal against E. coli and S. aureus. These results show that quaternized chitosan derivatives have great potential to be used as virucidal coatings, helping to prevent the spread of SARS-Cov-2 and other pathogens on surfaces, which can even be useful for the quick development of solutions for future pandemics.
Graphene and chitosan innovative materials for water treatment: Review

Chitosan (CS) is a natural polymer that has been extensively studied due to its specific formation, high chemical resistance and better physical properties. Chitosan has received wide attention due to its high adsorption potential and low cost. Among the advantages of chitosan can bind to heavy metal ions thanks to its form which has hydroxyl and amine groups. However, it also has drawbacks, for example sensitivity to pH, low thermal stability and low mechanical strength, which make it possible to limit the application of chitosan in the field of waste water treatment. The combination of graphene and chitosan allows us to obtain new materials with new properties and with improved adsorption performance. In the following, the preparation of GO-CS fibres, hydrogel and aerogel, and the applications of GO-CS nanocomposites are discussed. Also discusses the results obtained by researchers regarding the ability of the hydrogel to adsorb various contaminants, including cationic and anionic dyes and heavy metals.
Chemical modifications in the structure of marine polysaccharide as serviceable food processing and preservation assistant: A review

Now some marine polysaccharides, such as chitosan, sodium alginate and agar, have been proven to exhibit antibacterial, antioxidant functions and biocompatibility, which are often used to preserve food or improve the physicochemical properties of food. However, they still have the defects of unsatisfactory preservation effect and biological activity, which can be remedied by its modification. Chemical modification is the most effective of all modification methods. The advances in common chemical modification methods of chitosan, sodium alginate, agar and other marine polysaccharides and research progress of modified products in food processing and preservation were summarized, and the influence of additional reaction conditions on the existence of chemical modification sites of polysaccharides was discussed. The modification of functional groups in natural marine polysaccharides leads to the change of molecular structure, which can improve the physical, chemical and biological properties of marine polysaccharides. Chemically modified products have been used in various fields of food applications, such as food preservatives, food additives, food packaging, and food processing aids. In general, chemical modification has excellent potential for food processing and preservation, which can improve the function of marine polysaccharides.
