First, SHBG causes an increase of intracellular cyclic adenosine monophosphate (cAMP) [7, 8]; second, cAMP targets protein kinase A (PKA) [9]; third, increased PKA activity counteracts estradiol-induced extracellular signal-regulated kinase (ERK) activation [10], and inhibition of the anti-apoptotic effects of estradiol [9], reducing estrogen receptor α (ERα) transcription efficiency [11]. The final effect of SHBG in breast cancer cells is inhibition of cell proliferation and apoptosis induction [10, 12].
