2012, Comparative Biochemistry and Physiology - B Biochemistry and Molecular BiologyCitation Excerpt :Considering reports in the literature on the isolation of toxins from Anthothoe chilensis (Lesson, 1830) specie (Retuerto et al., 2007), this is the first basic phospholipase A2 isolated and characterized from this species. Amino acid residues at positions 6–12 (in particular W10, G12) are proposed to form the Ca2 +-binding segment, similar to that found in other group III secretory PLA2s from the lizards (Sosa et al., 1986), bee venoms (Nakashima et al., 2004) and humans (Valentin and Lambeau, 2000). The amino acid residues corresponding to positions 19–20 correspond to the catalytic site, represented by H(23) and D(24), in both PLA2 (Anch TX-I and II), corresponding to H(48) and D(49) in the enzymatically active pancreatic PLA2.

2010, ToxiconCitation Excerpt :Interestingly, similar to PLA2s of high enzymatic activity, TI-Nh in the concentration range 1–10 μM evokes differentiation of rat pheochromocytoma PC12 cells into neuron-like cells. Although there are some data to indicate that PLA2 enzymatic activity may be involved in the induction of cell differentiation (Nakashima et al., 2004), the mechanism of cell differentiation evoked by PLA2s is not completely clear. The fact that TI-Nh with low lipolytic activity induces cell differentiation as effectively as PLA2s with high enzymatic activity may indicate that the mechanism is independent of the enzymatic activity.

2009, ToxiconCitation Excerpt :The overall three dimensional structure of bee venom group III PLA2 reveals the striking features of the PLA2 fold, which includes a well-defined Ca2+-loop, three large α-helices and a β-wing-like structure. Thus, BcPLA2 may have the same molecular properties as those of bee (Rizzo et al., 2000; Nakashima et al., 2003, 2004) and lizard PLA2 (Sosa et al., 1986). In the present study, we found that BcPLA21 stimulated insulin secretion only in conditions of high glucose concentration.
