Abstract

We have shown that manassantin A downregulated the HIF-1α expression and inhibited the secretion of VEGF. We have also demonstrated that the 2,3-cis-3,4-trans-4,5-cis-configuration of the tetrahydrofuran is critical to the HIF-1 inhibition of manassantin A.

Apigenin promotes the degradation of HIF-1α through HSP90 inhibition [16]. Manassantin A exhibits the activity of downregulating HIF-1α protein expression (IC50 = 1 ~ 10 nM) and inhibiting VEGF secretion [29]. Berberine enhances proteolysis of HIF-1α by proteasome [30].

The anti-inflammatory activity of manassantins is mediated by its inhibition of NF-κB transactivation activity [503]. Manassantin A and B have been considered as potent HIF inhibitors because manassantin A and B exert their inhibitory activity against the hypoxia-induced HIF-1α and VEGF expression at the concentration range of 1 nM to 1 μM, and a complete inhibition was observed at 100 nM in T47D human breast tumor cells [504,505]. However, the precise molecular mechanism has not been fully elucidated, suggesting further in-depth investigation especially in models with tumor hypoxic microenvironment.

A variety of HIF-1 inhibitors also have been identified in cell-based screens from natural product libraries [95] and activity-guided fractionation using plants and marine organisms, including sodwanone and yardenone triterpenes from the marine sponge Axinella [96], manassantin B and 4-O-demethylmanassantin B from Saururus cernuus [97], laurenditerpenol, a new diterpene from the tropical marine alga Laurencia intricata [98], and the terpenoid tetrahydroidoquinoline alkaloids emetine, klugine, and isocephaeline [99]. In 2009, Hong and co-workers reported manassantin A (35) and analogs with modified tetrahydrofuran configurations as HIF-1 inhibitors [100]. The manassantin A showed downregulation of the HIF-1α expression (IC50 = 1–10 nM) and inhibition of the secretion of VEGF.
