Manassantin A and B have been considered as potent HIF inhibitors because manassantin A and B exert their inhibitory activity against the hypoxia-induced HIF-1α and VEGF expression at the concentration range of 1 nM to 1 μM, and a complete inhibition was observed at 100 nM in T47D human breast tumor cells [504,505]. However, the precise molecular mechanism has not been fully elucidated, suggesting further in-depth investigation especially in models with tumor hypoxic microenvironment.

In 2009, Hong and co-workers reported manassantin A (35) and analogs with modified tetrahydrofuran configurations as HIF-1 inhibitors [100]. The manassantin A showed downregulation of the HIF-1α expression (IC50 = 1–10 nM) and inhibition of the secretion of VEGF.
