Materials and methods
Preparation of the total RNA. Live individuals of adult P. fucata were obtained from the Guofa Pearl Farm in Beihai, Guangxi Province, China. Total RNA was extracted from the mantle tissue with the RNAzol RNA isolation kit, (Biotecx, Houston, TX, USA) according to the manufacturer’s instructions. RNA integrity was determined by separation on a 1.2% formaldehyde-denatured agarose gel and stained with ethidium bromide. The quantity of RNA was determined by measuring OD260nm with an Ultrospec 3000
Isolation and sequence analysis of cDNA sequence encoding oyster tyrosinase
A cDNA product of 408 bp was obtained by RT-PCR with two degenerate oligonucleotides (DP1 and DP2) using total mantle RNA as the template. Based on the sequence, two gene-specific primers were synthesized for 5′-RACE (5RP) and 3′-RACE (3RP), respectively, and were used to amplify the 5′ and 3′ nucleotide sequence of putative oyster tyrosinase cDNA by touchdown PCR.
