Xyloglucan nonasaccharide, a naturally-occurring oligosaccharin, arises in vivo by polysaccharide breakdown

Summary

The production of xyloglucan nonasaccharide (XG9), an oligosaccharin that antagonises the growthpromoting action of auxin and H+, was investigated in vivo. Cell suspension cultures of spinach (Spinacia oleracea L.) were fed L[3H]fucose and the appearance of [3H]oligosaccharides and [3H]polysaccharides in the culture medium was monitored chromatographically. Two [3H]oligosaccharides accumulated in the medium — XG9 and an acylated derivative of XG9. After a short lag (∼0.5h), soluble [3H]polymers were released into the medium at a constant rate. In contrast, [3H]XG9 accumulated in the medium at a steadily accelerating rate for ∼5-8h. The data suggest a precursor→product relationship between [3H]polysaccharide and [3H]XG9. We conclude that XG9 arises by the partial hydrolysis of pre-formed xyloglucan.
