Abstract

Induction of allergic and autoimmune reactions by drugs and other chemicals constitutes a major public health problem. Elucidation of the underlying mechanisms might help improve diagnostic tools and therapeutic approaches. Here, Peter Griem and colleagues focus on several aspects of neoantigen formation by xenobiotics: metabolism of xenobiotics into reactive, haptenic metabolites; polymorphisms of metabolizing enzymes; induction of costimulatory signals; and sensitization of T cells.

Metals, autoimmunity, and neuroendocrinology: Is there a connection?

It has been demonstrated that metals can induce autoimmunity. However, few studies have attempted to assess and elucidate the underlying mechanisms of action. Recent research has tried to evaluate the possible interactions of the immune system with metal ions, particularly with heavy metals. Research indicates that metals have the potential to induce or promote the development of autoimmunity in humans. Metal-induced inflammation may dysregulate the hypothalamic-pituitary-adrenal (HPA) axis and thus contribute to fatigue and other non-specific symptoms characterizing disorders related to autoimmune diseases. The toxic effects of several metals are also mediated through free radical formation, cell membrane disturbance, or enzyme inhibition. There are worldwide increases in environmental metal pollution. It is therefore critical that studies on the role of metals in autoimmunity, and neuroendocrine disorders, including effects on the developing immune system and brain and the genetic susceptibility are performed. These studies can lead to efficient preventive strategies and improved therapeutic approaches. In this review, we have retrieved and commented on studies that evaluated the effects of metal toxicity on immune and endocrine-related pathways. This review aims to increase awareness of metals as factors in the onset and progression of autoimmune and neuroendocrine disorders.
Drug-induced lupus
2020, Systemic Lupus Erythematosus: Basic, Applied and Clinical Aspects
Drug-induced lupus (DIL) is a side effect of long-term use of up to 100 medications. Rheumatologic symptoms including fever, malaise, weight loss, polyarticular arthralgias, and symmetric myalgias develop in less than 1% of treated patients for some and less than 0.1% for most of the drugs with propensity to induce lupus-like symptoms. However, autoantibody induction by some drugs is considerably more common, and autoantibody serology can be indistinguishable from that of systemic lupus erythematosus. The defining, although retrospective, feature of DIL is resolution of symptoms after discontinuation of the suspected medication, usually without the need for immunosuppressive agents. Several genetic factors predispose to DIL, but dose and duration of therapy are the predominate risks. It is unlikely that the ingested, parent compound mediates DIL, and reactive metabolites of multiple pharmacological classes of lupus-inducing drugs are candidate initiators. Experimental studies support several possible mechanisms to account for autoimmunity in DIL, including specific and nonspecific activation of immune cells, cytotoxicity-mediated release of autoantigens, and the disruption of immune tolerance machinery. From the basic science perspective, DIL is a fascinating window into how the immune system normally acquires self-tolerance and how these mechanisms can fail, resulting in the development of autoimmunity and lupus-like disease.

Models of autoimmunity fall into four categories: (a) those induced by immunization with self-antigen, (b) those induced by exogenous agents, (c) those which arise spontaneously, and (d) those which are produced by genetic manipulation. The autoimmunity exhibited by these models covers a spectrum of diseases which fall into the two broad categories, organ-specific and systemic autoimmunity. Animal models of autoimmune diseases have played an essential role in the discovery of many of mechanisms that result in the breaking of self-tolerance. This chapter describes a number of experimental animal models of autoimmunity and the underlying mechanisms that lead to disease.
Recent advancements in the mitigation of obnoxious nitrogenous gases

Nitrogenous gaseous emissions commonly have an obnoxious odor associated with it, which when discharged into the environment results in serious environmental problems and health hazards. Several strategies for mitigation of nitrogenous odorants have been reported which include physical, chemical and biological methods. Biological treatments are widely employed because of their efficiency even at low concentration, where physical and chemical methods are not effective. Most commonly used biological treatment methods are biofiltration, biotrickling filters and membrane bioreactors with innovative reactor design, mixing pattern, and air sparging, for example FEBR, ALR, etc. These treatment methods require a critical assessment for the mitigation of obnoxious nitrogen emissions, especially in the context of environmental protection. This review offers a critical evaluation of treatment methods for the mitigation of nitrogenous odorous compound with a key emphasis on biological treatment systems. Also, various mathematical modelling techniques required for optimized operation of biotreatment systems has been discussed.
Comparison of in vivo immunomodulatory effects of 5-hydroxymethylfurfural and 5, 5'-oxydimethylenebis (2-furfural)

The standard of 5-Hydroxymethylfurfural (5-HMF) existed in dextrose injection as an inevitable by-product during high-temperature setrilization has been included in pharmacopoeias considering its hazardous effects on human health. We found that the concentrations of 5-HMF in some traditional Chinese medicine injections (TCMIs) far exceeded its limit in dextrose injection. Besides, we detected 5, 5′-Oxydimethylenebis (2-furfural) (OMBF) in those TCMIs containing high concentrations of 5-HMF. We investigated the in vivo immunomodulatory effects of 5-HMF and OMBF at three dose levels using the reporter antigen popliteal lymph node assay (RA-PLNA), which allows the straightforward examination and mechanistic study of immunotoxicity of low molecular weight compounds. We found that 5-HMF increased the production of IgG_2a and IFN-γ when co-injected with TNP-OVA, indicating its capability of providing a co-stimulatory signal to evoke a typical type-1 immune response. Compared with the 5-HMF, OMBF elevated the production of IgG₁, IgG₂, IL-4 and IFN-γ in response to both reporter antigens, suggesting that OMBF can act as a neo-antigen or neo-epitope to elicit a mixed type-1 and type-2 immune response. It indicates that both 5-HMF and OMBF have immunosensitizing potential with different mechanisms, and exposure to 5-HMF and OMBF may represent a safety concern for humans.
