Alcian blue staining of proteoglycans in polyacrylamide gels using the “Critical electrolyte concentration” approach

Abstract

A method is described here for Alcian blue staining of proteoglycans in polyacrylamide gels; this is illustrated using extracts obtained from bovine corneal stroma. Other available methods for visualization of proteoglycans can produce nonspecific staining and frequently a high background in the gel; with a “critical electrolyte concentration” approach, specific staining against a clear background can be obtained.

    This new fraction was referred as the LS-AIM (Laemmli-soluble, acetic acid insoluble).     Samples were loaded on gels, which were subsequently run for 15 min at 100 V, then for about 1 h at 150 V. For visualization of proteins, gels were stained with Bio-Safe Coomassie G250 (Bio-Rad, ref. 161-0787) and with silver nitrate according to the protocol of Morrissey (1981).     To stop the process, solution II was decanted and solution III (50% (v/v) methanol, 12% (v/v) acetic acid) was immediately added (Blum et al., 1987). Others gels were submitted to Alcian blue method for GAGs detection, according Wall and Gyi (1988).
