Summary

Spectrophotometric and fluorometric direct evaluation of thin-layer chromatograms is described. Scanning can be carried out by determination of spectral reflectance, fluorescence and fluorescence quenching. The techniques described are suitable for characterization and identification of microgram amounts of substances by directly plotting their reflectance or fluorescence spectra. Several examples (amino acids, DNP-, DANS-, PTH-amino-acids, nucleic acid derivatives) are given, influencing factors and the reproducibility are discussed.

Zusammenfassung

Die spectrophotometrische und fluorometrische Direktauswertung von Dünnschicht-Chromatogrammen wird beschrieben. Quantitative Auswertungen können durchgeführt werden durch Messung der Remission, Fluoreszenz und der Fluoreszenzlöschung. Die beschriebenen Techniken eignen sich zur Charakterisierung und Identifizierung von Mikrogramm-Mengen von Substanzen, durch direkte Aufnahme ihrer Remissions- oder Fluoreszenzspektren. Zahlreiche Beispiele (Aminosäuren, DNP-, DANS-, PTH-Aminosäuren, Nukleinsäure-Derivate) werden angegeben, Einflußfaktoren und die Reproduzierbarkeit werden diskutiert.

On décrit l'évaluation directe, par spectrophotométrie et par fluorométrie, de chromatogrammes sur couche mince. Cette évaluation peut se faire par détermination soit de la réflexion spectrale, soit de la fluorescence, soit de l'extinction de la fluorescence. Les techniques décrites se prètent à la caractérisation et identification de substances, dans la région des microgrammes, par enregistrement direct de leurs spectres de reflexion ou de fluorescence. Plusieurs exemples (amino-acides, DNP-, DANS-, PTH-amino-acides, dérivés d'acides nucléiques) sont donnés, et sont discutés les différents paramètres ainsi que la reproductibilité.
