Zusammenfassung
Die LD50 bei Mäusen als Virulenzmaß für Ps. aeruginosa wird an einer Stichprobe von 20 Stämmen auf ihre methodische Zuverlässigkeit geprüft. Die ermittelten Werte ergeben eine eingipfelige, offenbar logarithmisch normale Verteilung. Das tierexperimentelle Modell ermöglicht quantitative Aussagen über die dosisabhängige Schutzwirkung von spezifischen Antikörpern gegen Pseudomonasbakterien, über die therapeutischen Effekte abgestufter Carbenicillin-Konzentrationen und die Kombination beider protektiver Maßnahmen. Der Synergismus zwischen der antibakteriellen Hemmstoffwirkung und der bei passiver Immunisierung durch zusätzliche Opsonisierung der Erreger gesteigerten Phagozytose ist über alle Konzentrationsbereiche additiver Natur. Es ergibt sich demnach kein Anhalt für die hypothetische Virulenzminderung von Pseudomonasbakterien, die unter subinhibitorischen Carbenicillin-Konzentrationen in der Zellwandsynthese gestört und trotz erheblicher Deformation noch vermehrungsfähig sind.
Summary
The LD50 of Ps. aeruginosa bacteria administered intraperitoneally to mice was studied for its reliability as an estimate of virulence. Its distribution in a sample of 20 strains could be shown to be logarithmically normal. Evidence as to the dose-dependent protection by strain and group specific antibody, the therapeutic effects of different carbenicillin concentrations, and the combination of both activities was gained by use of the LD50 model. The synergism resulting from the simultaneous administration of antiserum, which enhances phagocytosis by additional opsonization of the bacteria, and the antibiotic in different doses has proved to be merely additive. No particular therapeutic advantage can therefore be deduced from the observation that Pseudomonas bacteria are disturbed in their cell wall synthesis by subinhibitory carbenicillin concentrations.
