Abstract
In 6 dogs with gastric fistùla, burimamide, a histamine analog which blocks H-2 histamine receptors, was given in a dose of 500 μg/kg-hr as a background to step-dose responses with histamine (2–100 μg/kg-hr) or with betazole (50–5000 μg/kg-hr). The drug did not change the histamineresponsive H, electrolyte, or pepsin secretion, but did competitively inhibit both H+ and electrolyte responses to betazole, and reduced pepsin secretion by 90%. Three of the dogs in studies using pentagastrin (0.1–5.0 μg/kg-hr) had marked reduction in pepsin output but no effect on H+ or electrolytes by burimamide. With 2-DG stimulation in 3 dogs (1 dose, 100 mg/kg), burimamide had no effect on H+ or pepsin secretion. Larger doses of burimamide, 1–5 mg/kg-hr, produced side effects, especially retching and diarrhea, which precluded studies of these doses.
