Summary
Sephadex® gel filtration has been employed for the quantitative determination of the binding of3H-labeIled diisopropyl-phosphoro-fluoridate (DFP) in guinea-pig plasma.

The amount of DFP bound to the plasma proteins depends upon the plasma concentration of DFP; at 10−7 M more than 97%; at 10−6 M about 50% and at 10−4 M less than 2% is bound.

The uptake of DFP by erythrocytes was measured for two extreme DFP concentrations in blood. The amount of free, non-protein-bound substance was taken as a base for the calculation of the “true” erythrocytes-plasma ratios. In accordance with general pharmacokinetic experience, the uptake represented by the “true” T/M ratios was relatively higher at lower DFP blood concentration.

Quantitatively, the interaction with albumin seems to be the most significant process. The affinity constantK 01 =1.4×105M−1 was evaluated by means of a graphic extrapolation procedure. Since a sharply curved line was obtained, it is postulated that the characteristics of the binding sites of the albumin molecule would be influenced by particularly elevated concentrations of DFP. A change in tertiary structure of the albumin molecule is discussed as a possible explanation of this finding.
Zusammenfassung
Mit Hilfe der Sephadex®-Gel-Filtrationsmethode wurde die Bindung von3H-markierten Di-isopropyl-phosphorofluoridat (DFP) an Meerschweinchen-Plasma quantitativ bestimmt.
Es ergab sich:

Die Plasmabindung von DFP war von der DFP-Konzentration im Plasma abhängig, bei einer DFP-Konzentration von 1×10−7m waren mehr als 97%, bei 1×10−6m ungefähr 50% und bei 1×10−4m weniger als 2% eiweißgebunden.

Die Aufnahme von DFP in Erythrocyten wurde für zwei extreme DFP-Konzentrationen gemessen. Auf der Grundlage des freien, nicht plasmagebundenen Anteils wurden die „wahren“ Erythrocyten-Plasma-Quotienten errechnet. Wie für ein absättigbares Kompartiment zu erwarten war, lagen die wahren Erythrocyten/Plasma-Quotienten höher bei geringerer Plasma-Konzentration.

Im Blut spielt die Bindung von DFP an Albumin quantitativ die größte Rolle. Die Affinitätskonstante K 01 =1,4×105 mol−1 wurde graphisch durch Extrapolation ermittelt. Aufgrund des Kurvenverlaufs wird gefordert, daß die Eigenschaften der DFP-Bindungsstellen sich durch hohe DFP-Konzentration beeinflussen lassen. Als Ursache dieser Änderung der Bindungseigenschaften wird eine Störung der Tertiär-Struktur der bindenden Moleküle diskuttiert.
