Summary
Amino acid utilization by various strains ofBordetella pertussis, conditions for the preparation of casein hydrolysates, and the significance of pH for growth were studied. For achieving maximum growth the presence of casein hydrolysate amino acids in sufficient concentration is important since they determine total growth. The amino acids can be divided into two groups. The first group comprizes those which are at the same time sources of energy and whose utilization from the medium stops multiplication. Amino acids from the second group are utilized slowly and remain in the medium after growth cessation. The growth rate depends on the presence of all amino acids from the first group. They can be mutually substituted but the growth rate decreases.
The decisive factor for the quality of casein hydrolysate is its purification. The technique of splitting and the quality of casein are of secondary importance. The optimum pH for the growth of all field strains used lies between 7.4–7.9. Intensive aeration in submersed cultivation influences the pH level favourably, as under these conditions ammonia is removed from the medium.
