Zusammenfassung
Mit Vesicularstomatitis-Virus infizierte Chorioallantoismembranen von Hühnerembryonen wurden ultrahistologisch untersucht.
Nach Impfung in die Allantoishöhle ließen sich runde und stäbchenförmige Partikel, von denen angenommen wird, daß sie zum System des Vesicularstomatitis-Virus gehören, zuerst in den Zellen und Zellzwischenräumen der Chorionschicht nachweisen.
An das Mesoderm grenzende Randzonen von Chorion- und Allantoisschichtzellen, welche durch eine Membran vom übrigen Zytoplasma abgetrennt sind, enthalten ein granuliertes Material mit darin eingebetteten Viruspartikeln. Es wird angenommen, daß diese Regionen Bildungszentren der Virusteilchen sind.
Erst in späten Stadien, wenn die Allantoisschichtzellen bis auf die Zellmembranen zerstört sind, werden die Viruspartikel auch an den der Allantoishöhle zugewandten Oberflächen der entodermalen Zellen beobachtet.
Mit Hilfe des Negativ-Färbeverfahrens mit Phosphorwolframsäure konnte nachgewiesen werden, daß die stäbchenförmigen Partikel an einer Schmalseite abgerundet sind und an der anderen einen geraden Abschluß haben. Vereinzelt wurden Stäbchen mit Innenkörpern beobachtet. Die Mehrzahl der runden Teilchen ist mit einem schwanzförmigen Fortsatz ausgerüstet.
Summary
An ultra-histological investigation of chorio-allantoic membranes of chick embryos infected with stomatitis vesicularis virus has been carried out.
After inoculation into the allantoic cavity spherical and rod shaped particles, presumably belonging to the virus system of vesicular stomatitis, could be demonstrated. They appeared first in the cells and intracellular spaces of the chorion layer.
Granular areas bordering the mesoderm were observed in cells of the choionic and allantoic layers. These areas, which are separated from the remaining cytoplasma by a membrane, contain embedded virus particles. It is assumed that these areas represent the centres of virus development.
At a later stage, when the internal structure of the cells of the allantoic layer is destroyed, leaving the cell wall intact, the virus particles are observed also at the surface of entodermal cells facing the allantoic cavity.
It could be demonstrated by negative staining with phosphotungstic acid that the rod shaped particles have each a rounded and a square end. Single rods containing one inclusion were observed. The majority of the spherical particles were noted to possess tail-like appendages.
