Abstract
Cell cycle activity is a prerequisite for growth and development. In germinating seeds, cell cycle activity has been shown to precede visible germination. The paper reviews cell cycle research with dormant and germinating tomato seeds. Nuclear DNA replication, as envisioned by flow cytometry, starts in the embryonic root tip and precedes visible germination. This DNA replication also occurs during priming of seeds and in the presence of abscisic acid, despite the inhibition of germination due to both these treatments. In dormant and gibberellin deficient tomato seeds DNA replication is blocked. Immunological detection of ß-tubulin levels, showed that accumulation of this cytoskeletal protein is correlated with the onset of nuclear DNA replication. The studies with dormant and gibberellin deficient tomato seeds give strong indications for the occurrence of a G0 cell cycle phase in plants. The synchronization of the cell cycle in dry seeds, makes it an ideal material to study the regulation of growth and the cell cycle in plants. On the other hand, the analysis of cell cycle in germinating seeds will offer opportunities to get a more detailed knowledge of pre-germination seed development and its relation to seed quality.
