Comparison of Two Extraction Methods to Obtain Quality Genomic DNA from Eggplants (Solanum sp.)

Abstract

Eggplant (Solanum sp.) is the third most widely grown solanaceous vegetable after potatoes and tomatoes. The crop is characterized by the presence of high secondary metabolites, starch, and polysaccharide content, which interferes with the genomic DNA during extraction. It becomes necessary to reduce the effects of these interfering compounds to a low level by optimizing the extraction protocols. Different methods described so far have explored the leaves of eggplant for the isolation of genomic DNA. The DNA extraction method in this study explored eggplant seeds to isolate genomic DNA from eggplant. Here, we compare two different DNA extraction methods including CTAB (cetyltrimethyl ammonium bromide) and SDS (sodium dodecyl sulfate) with minor modifications using 15 seed accessions of eggplant. The results indicated significant differences in DNA contents between the two extraction methods (p < 0.05). The mean DNA yield from the 15 seed samples ranged from 645.6 ± 156.5 ng/μl for SDS methods and 824.5 ± 101.3 ng/μl for CTAB methods. The isolated DNA’s average spectrophotometric values (A260/280) were 1.52±0.03 and 1.86±0.04 for CTAB and SDS methods, respectively. The two methods generated good-quality genomic DNA useful for further genomic analysis and therefore recommended for Solanum and related species for DNA isolation.
