Abstract
Retroviral vector is the most commonly used vector for gene therapy. However, its use is limited because of the low titer. To overcome this problem, we improved the packaging cell line and concentrated the viral supernatant by the low-speed centrifugation and obtained retroviral titers up to 1012cfu/ml. This high titer preparation enabled us to transduce foreign genes efficiently into the neural cells, which are generally resistant to the transduction. It was also possible to massively transduce RSV- glioma cells transplanted into the mouse brain in vivo. This study showed that high-titer retrovirus was able to overcome the limitation of infectivity to the nervous system both in vitro and in vivo.
