Ziel
Die Rolle von Metallverbindungen in der Ätiologie von Malignomen der Kopfspeicheldrüsen wurde untersucht.
Methode
Makroskopisch gesundes Speicheldrüsengewebe von 46 Patienten wurde bei operativen Eingriffen gewonnen und zu Einzelzellen isoliert. Die Inkubation erfolgte mit Na2Cr2O7, NiSO4, CdSO4, ZnCl2 und Ethanol sowie mit Na2Cr2O7 in Kombination mit den anderen Substanzen. Der Einfluss von CdSO4 wurde durch Inkubationen mit Na2Cr2O7 und anschließende DNA-Reparaturintervalle mit und ohne Cadmiumzusatz untersucht. Zur Darstellung von DNA-Schädigung und -Reparatur wurde der Comet Assay eingesetzt. Die Auswertung erfolgte fluoreszenzmikroskopisch mit digitaler Bildverarbeitung.
Ergebnisse
Lediglich Na2Cr2O7 induzierte eine Erhöhung der DNA-Migration in Speicheldrüsenzellen. Die Inkubationen von Na2Cr2O7 mit NiSO4, ZnCl2 und Ethanol ergaben keine Erhöhung der allein durch Na2Cr2O7 hervorgerufenen Genotoxizität. Cadmium hatte jedoch einen signifikanten additiven Effekt: eine Hemmung der Reparatur der durch Na2Cr2O7 hervorgerufenen DNA-Schäden.
Fazit
Na2Cr2O7 wirkt potenziell karzinogen auf humane Speicheldrüsenzellen. CdSO4 zeigte keine direkten genotoxischen Auswirkungen auf Speicheldrüsenzellen, hemmte jedoch die DNA-Reparatur oxidativer DNA-Fragmentierung.
Abstract
Background
The etiology of malignomas of human salivary glands is examined.
Material and methods
Macroscopic, healthy salivary gland tissue from 46 donors was harvested during surgery. Single cells were isolated by enzymatic digestion. These were then incubated for 60 min with Na2Cr2O7, NiSO4, CdSO4, ZnCl2 and ethanol. Additionally, incubation with Na2Cr2O7 was combined with NiSO4, CdSO4, ZnCl2 and ethanol. The influence of CdSO4 was analyzed by altered combinations with Na2Cr2O7 during incubation and by the DNA-repair period. Evaluation was performed using fluorescent staining and digital analysis.
Results
Of all of the substances tested, only Na2Cr2O7 showed genotoxic effects. NiSO4, ZnCl2 and ethanol had neither genotoxic nor cofactorial impacts. CdSO4, however, caused additional genotoxic effects in combination with Na2Cr2O7, although it lacked direct genotoxic potential. A reduction of DNA-repair of Na2Cr2O7-induced oxidative damage by CdSO4 could be demonstrated.
Conclusions
In this investigation, sodium dichromate was identified as genotoxic in association with human salivary gland tissue. These effects could be increased by CdSO4, reinforcing DNA damage based on oxidative stress.
