Genetic Organization of Antibiotic Resistance Genes (aac(6′)-lb, aadA, and oxa9) in the Multiresistance Transposon Tn1331

Abstract

The multiresistance transposon Tn1331 has two 520 bp direct repeats flanking a DNA region similar to the variable portion of the integrons. This region harbors three antibiotic resistance genes, aac(6′)-lb, aadA, and a β-lactamase gene which encodes an enzyme that was characterized as an oxacillinase-carbenicillinase and was named OXA-9. Within the direct repeat located upstream of these genes there is a copy of the TEM β-lactamase promoter. Determination of ampicillin resistance levels of Escherichia coli harboring various recombinant clones and an insertion mutant showed that all three genes are transcribed from this promoter in a polycistronic mRNA. These results also indicated that the oxa9 gene possesses another functional promoter, located immediately upstream of this gene. The transcription start site of the mRNA driven by this promoter was identified by primer extension. Within the DNA sequence harboring all three antibiotic resistance genes it is possible to distinguish insert units as defined before in the variable region of the integrons (R. Hall, D. Brookes, and H. W. Stokes (1991) Mol. Microbiol. 5, 1941-1959).

    The differences between these two plasmids are based on the resistance genes present next to the blaTEM-1A gene. Indeed, pKp145-11b harbored a Tn1331-like element that carried, in addition to blaTEM-1A gene, three other resistance genes blaOXA-9 gene, ant(3’)-Ia gene and aac(6’)-Ib gene [26], as previously described [27]. Finally, as reported, pKp145-11b had high nucleotide sequence identity with plasmid pKp41M (accession number KY781949), another IncM plasmid carried by a K. pneumoniae isolate recovered from a parrot in Brazil [26].

    At the 3′ end of the gene, instead of the usual attC site, there is a copy of attI1*, which may have been formed by an illegitimate recombination event between the attC site located 3′ of aadA1 of an integron and the attI1 locus located 5′ of blaOXA-9 of another integron in which the blaOXA-9 gene cassette is adjacent to the 5′-conserved sequence (Sarno et al., 2002). The resulting structure defines a gene cassette consisting of aadA1-attI1* but that lacks the usual attC site, and another gene cassette that includes two genes aadA1-attI1*-blaOXA-9–attC (see Fig. 2A) (Ramirez et al., 2008; Sarno et al., 2002; Tolmasky, 1990; Tolmasky and Crosa, 1993). While the aadA1-attI1* gene cassette is excised by the IntI1 integrase at a very low frequency the gene cassette that includes both genes is fully functional (Ramirez et al., 2008).

    Also of interest was the detection of OXA-9-positive E. coli, which also carried aac(6′)-Ib and aadA1-like gene. A region within a Tn1331 transposon encoding blaOXA-9, aac(6′)-Ib and aadA-like genes was first identified in a clinical isolate of Klebsiella pneumoniae [19]. Therefore, the array may also be useful for detecting known linkages between genes.
