In adults, strong associations have been validated between the APOE genotype and Alzheimer disease, as well as recovery after traumatic brain injury (TBI).20,25,26 In adults, the APOE ε4 allele is associated with an increased risk of Alzheimer disease and worse recovery after TBI.

Additional indirect evidence linking apoE, GSK3, and Tau comes from a recent interesting gene expression study showing that apoE and GSK3 are both differentially expressed in the entorhinal cortex where Tau pathology is thought to begin (72).

The most prevalent variant of the APOE gene in the human population encodes the apoE3 protein, while other common variants that encode the apoE2 and apoE4 proteins are associated with disease (Mahley et al., 2006; Roses, 1997). A dose-dependent inheritance of the APOE4 allele is associated with increased risk for developing Alzheimer's disease (AD) (Corder et al., 1998; Roses, 1997) and poor outcome from neurological injury (Chapman et al., 2001; Corder et al., 1998; Grocott et al., 2001; Jordan et al., 1997; Kutner et al., 2000).
