2010, Biochemical and Biophysical Research CommunicationsCitation Excerpt :Treatment with a PKC inhibitor, Go6983, inhibited basal and UVB-induced sHB-EGF secretion (44.0 ± 16.6, p = 0.029) (Fig. 2A), suggesting the involvement of PKC in the basal and UVB-induced sHB-EGF secretion. Among the isoforms of PKC, PKCδ has been reported to be tyrosine-phosphorylated and activated by UV irradiation [13], so we examined the involvement of PKCδ in UVB-induced HB-EGF secretion. Expression of GαqQL increased UVB-induced phosphorylation of PKCδ, and treatment with Rottlerin, a PKCδ-specific inhibitor, inhibited UVB-induced activation of PKCδ and HB-EGF secretion from GαqQL-expressing cells from 226.0 ± 36.6 to 102.3 ± 24.0 (p = 0.029) (Fig. 2C), which indicates that PKCδ acts downstream of Gαq in mediating the UVB-induced secretion of sHB-EGF.

2010, Archives of Biochemistry and BiophysicsCitation Excerpt :Protein kinase C delta (PKCδ),1 originally discovered in mouse epidermis [1], has been extensively studied, and its roles in cell signaling as a pro- as well as an anti-apoptotic protein have been described [2,3]. Studies show that kinase-active PKCδ is involved in up-regulating cell apoptosis in response to various stimuli such as H2O2[4,5], Tumor Necrosis Factor-α (TNF-α) [6], UV radiation [7], and DNA-damaging agents [8]. Activation of PKCδ was found to be associated with inhibition of cell cycle progression [9].
