2022, The Lancet RheumatologyCitation Excerpt :IGFBP5 is a highly conserved and multifunctional secreted protein that binds to IGF and can have complex and varying effects on IGF signalling depending on the tissue type and context. In bone, IGFBP5 inhibits IGF-1 signalling36 by modulating binding to the IGF-1 receptor.30 Similarly, in mammary tissue, IGFBP5 regulates involution by inhibiting IGF-1 signalling.31
The role of insulin-like growth factors in modulating the activity of dental mesenchymal stem cells
2021, Archives of Oral BiologyCitation Excerpt :IGFBP-5 has been observed to enhance proliferation rates in dental MSCs in vivo, even under inflammatory conditions (Han et al., 2017). Interestingly IGFBP-5 has been found to inhibit proliferation in non-stem cell lines, such as in the case of malignancy (Dong, Zhang, Fang, & Liu, 2020; Kiefer et al., 1992; Schneider et al., 2001), whilst maintaining its proliferation-enhancing properties in other stem cell lines (Florini, Ewton, & Coolican, 1996), indicating it may have differential mechanisms of action in different cell types. IGFBP-5 has been observed to enhance the proliferation rate, migration, differentiation, and mineralisation of dental MSCs; interestingly, however, there is conflicting evidence as to the role of IGFBP-5 in non-dental MSC differentiation, with studies indicating it inhibits, rather than promotes, osteogenic differentiation (Han et al., 2017; Hao, Yang, Cao, Zhang, & Fan, 2020; Li et al., 2019; Liu, Wang, et al., 2015; Magnucki et al., 2013; Morsczeck et al., 2005; Mukherjee & Rotwein, 2008).
