Upon ligand binding, such as epidermal growth factor (EGF), EGFR forms a dimer and activates several downstream signal pathways to promote cell growth [6–8]. At the same time, EGFR itself needs to be tightly regulated through a variety of posttranslational modifications, and is then subjected to recycle, degradation, or nuclear localization [4,5].

In line with this also, phosphorylation of the EGFR residue Tyr1173, a known EGFR autophosphorylation target (32), became detectable within 5 min (Fig. 1). No phosphorylation of EGFR residue Tyr1045, which represents an EGFR internalization site (33, 34), was observed, suggestive of ligand (EGF)-independent EGFR activation.
