Although estrogen receptor expression in this cell line has been reported, no reports on its expression with passage number have yet been published.

Although TGF-β1 has been known to stimulate proliferation and differentiation of osteoblasts, it has the suppressive effect on the expression of osteocalcin (Chung et al., 1999). The gene expression of osteocalcin in PDL cells was also downregulated after 4 h culture with TGF-β1, and furthermore the extent of TGF-β1-induced suppression of osteocalcin mRNA was dose dependent from 0.032 to 10 ng/ml (Brady et al., 1998).

2011, BoneCitation Excerpt :These data directly contradict the results of Matsuguchi et al. and Liu et al., who recently reported that SP600125 decreased osteogenic potential in MC3T3-E1 cells [16,28]. It is difficult to reconcile the findings of this study with those of Matsuguchi and Liu, although one potential explanation was their use of MC3T3-E1 cells of unreported passage number; MC3T3-E1 cells can show reduced BMP-responsiveness at higher passages [29]. Further work with animal models will be required to resolve the in vivo effects of JNK inhibition on bone.
