GPCRs: The most promiscuous druggable receptor of the mankind
2021, Saudi Pharmaceutical JournalCitation Excerpt :The genes of the frizzled receptors were first isolated from drosophila in 1989, followed by a discovery in mammals (Vinson et al., 1989). In mammals, 10 receptor subtypes have been identified and named frizzled 1–10 (Koike et al., 1999, Wang et al., 1996). Three signaling pathways have been characterized by frizzled receptors.
FZD<inf>10</inf>-Gα<inf>13</inf> signalling axis points to a role of FZD<inf>10</inf> in CNS angiogenesis
2017, Cellular SignallingCitation Excerpt :The ten FZDs, which are generally activated by the 19 members of the WNT family of secreted lipoglycoproteins with unknown isoform selectivity, can be subdivided into four homology clusters grouping FZD1,2,7, FZD3,6, FZD4,9,10 and FZD5,8 [1,3]. Among the receptors in class F, one of the most enigmatic representatives is FZD10 because of its temporal and spatially restricted expression pattern during embryonic development and poorly understood signal transduction mechanisms [4–7]. Expression pattern analysis in the developing central nervous system (CNS), indicates an important but yet undefined role of FZD10 in early CNS development [5,6,8].
Frizzled Receptors in Development and Disease
2016, Current Topics in Developmental BiologyCitation Excerpt :A search for additional vertebrate Frizzled family members using degenerate PCR identified eight family members in mammals, as well as multiple family members in birds and fish (Wang et al., 1996). Two additional mammalian Frizzled genes were identified subsequently, bringing the total to 10 (Koike et al., 1999; Wang et al., 1997). The large number of vertebrate Frizzled genes together with their receptor-like structure led Wang et al. (1996) to suggest that “If Frizzled proteins act as receptors, that would imply the existence of a corresponding family of ligands.
The Role of Signaling Pathways in Cervical Cancer and Molecular Therapeutic Targets
2014, Archives of Medical ResearchCitation Excerpt :Another mechanism involved in the upstream activation of the Wnt/β-catenin pathway is the over-expression of activators such as Wnt ligands, frizzled receptors and disheveled (Dvl). In cervical cancer cell lines, the overexpression of Wnt10B-14 (158,159), Fzd-10 (160), and Dvl-1 (161) has been reported. In a genome expression analysis conducted in HPV-16-associated cervical cancer cases, a significant increase in Wnt-4, -8a, Fzd2, GDK3b and β-catenin compared with normal cervical epithelia was observed (162).
